Enhancement of solubility of Domperidone by using combination of solid dispersion and liquisolid techniques with natural and synthetic polymers

Domperidone is a dopamine antagonist widely used as an antiemetic and gastroprokinetic agent; however, its therapeutic effectiveness is limited due to its poor aqueous solubility and low oral bioavailability. The present study aimed to enhance the solubility and dissolution rate of domperidone by employing a combined solid dispersion and liquisolid technique. Solid dispersions of domperidone were prepared using suitable natural and synthetic hydrophilic carriers(Manihot esculenta starch,Aloe vera gel powder and methyl cellulose) through a solvent evaporation method to improve drug wettability and dispersion. The optimized solid dispersion(drug:carrier ratio 1:3 was further incorporated into a liquisolid system using non-volatile solvent(Tween80), carrier materials(Manihot starch,Aloe vera gel powder and Micro crystalline cellulose), and coating material(Talc) to enhance the flow properties and compressibility.
